Competitors will start outside Nationwide in Pipers Way at 11am.
Spokesman Bev Rawlings said it would be a special event for the town.
"From the weather forecasts I have seen it's not going to rain, which is good," she said.
Faces featured in the Adver in the build up to the event will be among the crowd These include Olympic marathon runner Matt O'Dowd and Patrick Broderick, of Ruskin Avenue, who is a keen supporter of good causes.
The Evening Advertiser has teamed up with Swindon Council and Nationwide to organise the race. And it will be the first time in 20 years a running event of this scale has been held in the town. All money raised will go to charity.
